Sender Based Routing IS possible with Exchange 2007 and 2010 with an addon tool called ptecs EXSBR. ExSBR is a Hub Transport Agent which extends routing possibilities in Exchange Server. It is able to route by sender's smtp domain name or AD group memberships.
You can get an eval version from me, if you want to. Just write an email to exsbr at ptecs dot de with the number of your mailboxes, servers and hub transport servers.

I fully understand you concern. However, as far as I know, for Exchange
2007 server, it will use different routing method like DNS or SmartHost
server to send outbound mails based on the REMOTE email domain that emails
are sent to, instead of the ACCEPTED email domains which emails are sending
from. I.E. Exchange server will NOT route the mails based on sender's
domain.
However, I am stilling researching for any possible work around. I will get
you posted here on this issue.
